Programma  COURSE CONTENT. The course is aimed at providing the students with the knowledge in advanced pharmaceutical technology and drug delivery; in particular the use of colloidal systems and the more recent micro and nano-technological platforms will be thoroughly discussed. The course will highlight how pre-formulation studies are paramount in dictating the quality of the final formulation and the choice of the proper administration route. Factors affecting the rate of dissolution and absorption through biological membranes of drugs will be detailed together with the aproaches to increase dissolution rates and the use of penetration enhancers. Colloidal systems in pharmaceutical technology: properties, stability issues, surface energy. Suspensions and emulsions in pharmaceutical technology: components, preparation methods, strategies to reduce instability. Use of biodegradable polymers in drug delivery and pharmaceutical technology. Preparation of polymeric and lipidic micro-nanoparticles for the administration of biotech drugs. Particular interest will be dedicated to the characterization techniques of the nanocarriers for drug delivery. Use of gold nanoparticles and super paramagnetic nanoparticles for diagnostic and drug delivery purposes. Role of pharmaceutical technology to improve the biological stability and performance of vaccines. Transdermal and transsmucosal routes for non conventional administrations of biotech drugs. Cyclodextrins as natural carriers for drugs and si-RNA delivery. Carbon nanotubes as innovative carrier for intracellular delivery of biotech drugs. Basic concepts of the good manufacturing practice. LEARNING OUTCOME The students will acquire 1. knowledge lying at interface between the production of biotech drug and the formulation of the drug itself; 2. the main colloidal and nanotechnological systems used for biotech drug delivery. Such systems represent the most promising approach for biotechnological drug formulation.

Stefano Salmaso is currently Assistant Professor of the Faculty of Pharmacy, at the University of Padua (Italy) where he serves as lecturer of “Pharmaceutical Technology” teaching to undergraduate students enrolled in the master course of “Pharmaceutical Biotechnology”. He received his M.S. degree in “Chemistry and Pharmaceutical Technology” from the University of Padua in 1998 defending a thesis with the title “Peptides with Anticoagulant Activity: Structure, Stability and Function of N-terminal fragments of Hirudin HM2” and Ph.D. in “Pharmaceutical Sciences” in 2004 from the University of Padua with a dissertation on “Specific anti-tumour targetable β-cyclodextrin-PEG-Folate drug delivery system: synthesis, physico-chemical characterization and in vitro activity”. In 2000 he was granted a fellowship at Department of Pharmaceutical Sciences of the University of Padova where he was investigator in a study aimed at developing “Macromolecular systems for drug delivery”. During his career, he completed his training in international awarded research groups. In 2002 he was visiting researcher at the School of Pharmacy, University of Paris-Sud XI in Patrick Couvreur’s research group and at I.B.M.C-University of Strasbourg where he investigated by cellular models new cyclodextrin derivatives for active targeting of anticancer drugs. In 2005 he was visiting scientist at the Department of Pharmaceutical Sciences, Northeastern University – Boston (USA) where he joined Prof. Vladimir Torchilin’s gourp for the development of “pH-sensitive carriers for sparingly soluble pharmaceuticals”. In 2008 he joined Prof. Vladimir Torchilin’s laboratory as main investigator at the “Center for Pharmaceutical Biotechnology and Nanomedicine”, Northeastern University - Boston (USA). He developed a new project aimed at studying “Targeted nanocarriers for brain delivery”.

Risultati dell'apprendimento previsti Gli obiettivi del corso sono di dare agli studenti i fondamenti della moderna tecnologia farmaceutica con particolare enfasi al supporto che essa svolge nella formulazione di farmaci di origine biotecnologica. Il corso in particolare verte su preparazione di sistemi farmaceutici di recente evoluzione quali microparticelle polimeriche, sistemi transdermici, formulazioni per vaccini.
